This question was partly based on an Application in 3.5 (Genetic modification and biotechnology) relating to gene transfer in bacteria using restriction endonucleases and DNA ligase. It was also based on an understanding of DNA replication from 2.7 (DNA replication, transcription and translation). About half of candidates answered the question correctly, but a moderate discrimination index shows that some otherwise well-prepared candidates did not realise that DNA polymerase makes covalent bonds between nucleotides.
